The proposed experiments focus on determining if this is a direct or indirect process. Such information could allow development of new strategies for both prevention and treatment of MR Staphylococci by targeting the RNR as a regulator of this important virulence factor. Specific aims include: 1) to determine the relationship between increased amino acid substrate specificity and the hypermutable phenotype of a S. aureus RNAII mutant, 2) to explore the impact of the MR regulator, MecA, on the regulatory properties of the ribonucleotide reductase of S. aureus, and 3) to identify the major substrate specificity differences in S. aureus RNR between wild type and RNAI mutant strains. The specific aims of this proposal will be achieved by a multidisciplinary approach that includes gene discovery, directed mutagenesis, recombinant DNA methods, in vitro transcription and translation, and in vivo RNAI mutant analysis. Thus, this proposal will provide not only a mechanistic understanding of how Staphylococcus aureus invades the human host, but will also contribute novel information on the molecular basis of this pathogenic strain’s greater susceptibility to [unreadable]-lactam therapy.The present invention relates generally to circuits for limiting the current in a load and, more particularly, to an improved current limiting circuit.
